Medora was founded in 1883 by a French nobleman who was married to a New York heiress. The Marquis de Mores named the townsite after his wife, Medora van Hoffman. The Marquis tried various different business schemes but was forced to admit defeat and left the area four years later.

The town was a center for the cattle trade and for some local mines. Following the intervention of Bismarck businessman Harold Schafer in the 1960s and the subsequent restoration projects, Medora has developed into one of the premier tourist destinations in North Dakota.

The area was also the home of Theodore Roosevelt in the 1880s and the town profits from its proximity to the national park named after him and the surrounding Badlands area.
